Overview
dexamethasone + orelabrutinib is an investigational combination therapy consisting of the selective Bruton's tyrosine kinase (BTK) inhibitor orelabrutinib and the synthetic glucocorticoid dexamethasone. It is currently being evaluated in Phase 3 clinical trials for the treatment of newly diagnosed primary immune thrombocytopenia (ITP). Orelabrutinib, developed by InnoCare Pharma, covalently binds to BTK, inhibiting B-cell receptor signaling and subsequent B-cell activation and autoantibody production. Dexamethasone acts as a glucocorticoid receptor agonist, providing rapid anti-inflammatory and immunosuppressive effects. The combination is designed to provide a synergistic effect by targeting both the underlying B-cell-mediated pathology and providing immediate immune modulation to increase platelet counts in patients with ITP.
