Overview

Dexamethasone acts primarily as an agonist of the glucocorticoid receptor, leading to broad anti-inflammatory, immunosuppressive, and anti-edematous effects; it downregulates inflammatory mediators and suppresses immune cell migration[1]. Oxymetazoline binds to and activates alpha-1A and alpha-2 adrenergic receptors on vascular smooth muscle, causing vasoconstriction and reduced mucosal edema, thereby relieving nasal congestion[2]. The combination is most commonly administered intranasally for the treatment of chronic rhinitis and nasal congestion, with evidence suggesting superior efficacy for symptom relief and prevention of rebound congestion when compared to either drug alone[6][7]. There is clinical research supporting the safety and efficacy of combined therapy, noting that corticosteroid addition may reduce the risk of rhinitis medicamentosa seen with prolonged oxymetazoline use[6][7].
