Dexamethasone + tretinoin is a combination of two active pharmaceutical ingredients—a synthetic glucocorticoid corticosteroid (dexamethasone) and a retinoid (tretinoin, also known as all-trans retinoic acid). This combination is used in both topical and systemic formulations for various indications. Topically, it may be compounded to treat dermatological conditions such as melasma, post-inflammatory hyperpigmentation, acne scars, age spots, and vitiligo. Dexamethasone acts primarily as an anti-inflammatory agent by binding to the glucocorticoid receptor and modulating gene expression to suppress inflammation. Tretinoin promotes cell turnover by activating nuclear retinoic acid receptors (RARs), leading to increased epidermal cell proliferation and differentiation. Systemically or orally, the combination has been studied for immune modulation in diseases like primary immune thrombocytopenia—where dexamethasone provides immunosuppression while tretinoin exerts additional immunomodulatory effects that promote thrombopoiesis[3][8].
