**Dexamethasone + venetoclax** is a combination of two pharmaceutical agents used primarily in clinical research and treatment for hematologic malignancies, most notably in **relapsed or refractory multiple myeloma (RRMM)**, particularly in patients with the t(11;14) chromosomal translocation. **Venetoclax** is a selective BCL-2 inhibitor, promoting apoptosis in cancer cells that are dependent on BCL-2 for survival. **Dexamethasone** is a synthetic glucocorticoid corticosteroid with broad anti-inflammatory and immunosuppressive actions and is commonly used as part of combination regimens for various cancers. The combination leverages the pro-apoptotic action of venetoclax in BCL-2 dependent myeloma cells and the anti-myeloma and supportive effects of dexamethasone. Recent studies compare this combination (often called "VenDex") to other standard therapies in RRMM with t(11;14), showing promising efficacy[10].
