Overview

dexmedetomidine + esketamine + ropivacaine is an investigational drug combination being evaluated by Peking University First Hospital for epidural labor analgesia. This combination therapy integrates three distinct pharmacological agents: dexmedetomidine, a selective alpha-2 adrenergic agonist that provides sedation and analgesia; esketamine, a non-competitive NMDA receptor antagonist that offers potent analgesic effects and prevents central sensitization; and ropivacaine, a long-acting amide local anesthetic that blocks sodium channels to inhibit nerve impulse conduction. The combination is designed to provide effective pain relief during labor while minimizing the adverse effects associated with traditional opioid-based regimens, such as sufentanil-ropivacaine, including pruritus and potential neonatal respiratory depression.
