DF1001 + nivolumab is an investigational combination therapy being evaluated in advanced solid tumors, notably those with HER2 expression. DF1001 is a first-in-class immune engager (TriNKET) developed by Dragonfly Therapeutics that targets human epidermal growth factor receptor 2 (HER2) on tumor cells and potently activates natural killer (NK) cells, γδ T cells, and CD8+ T cells through co-stimulation of NKG2D and CD16. Nivolumab is an anti-PD-1 monoclonal antibody that blocks the programmed death-1 (PD-1) pathway and enhances the immune system’s ability to recognize and destroy cancer cells. The combination is studied for synergistic effects in stimulating anti-tumor immunity in patients with advanced solid tumors, particularly those with HER2-positive, HER2-low, or otherwise treatment-resistant disease[1][2][3][4][5][7].
