Overview

DF6215 + pembrolizumab is an investigational combination therapy composed of DF6215, a half-life extended, monovalent recombinant human interleukin-2 (IL-2), and pembrolizumab, an anti-PD-1 monoclonal antibody marketed as KEYTRUDA. DF6215 is engineered to preferentially stimulate anti-tumor CD8+ T cells and NK cells through IL-2 receptor activation, especially IL-2Rα and increased IL-2Rβγ signaling, while reducing toxicities seen with historical IL-2 drugs, such as aldesleukin. An Fc fusion prolongs DF6215's half-life, allowing less frequent dosing. Pembrolizumab blocks PD-1, enhancing anti-tumor immune responses by preventing T cell inhibition. Preclinical studies showed DF6215 acts synergistically with PD-1 blockade, expanding effector immune cells and improving anti-tumor efficacy without inducing capillary leak or cytokine release syndromes. The combination is currently being evaluated in a first-in-human, multi-arm Phase 1/1b trial for advanced solid tumors, including advanced melanoma post anti-PD-1 and platinum-resistant ovarian cancer[1][2][3][5][7].
