DFF332 + everolimus is a combination therapy under investigation for the treatment of advanced or relapsed clear cell renal cell carcinoma (ccRCC) and other malignancies with HIF2α-stabilizing mutations. DFF332 is a small molecule inhibitor of hypoxia-inducible factor 2 alpha (HIF2α), a transcription factor implicated in tumor growth and progression in renal cell carcinoma and other cancers. Everolimus (RAD001) is an mTOR (mechanistic target of rapamycin) inhibitor used for antineoplastic therapy. Together, this regimen aims to target both the HIF pathway (key driver in ccRCC and certain genetic tumor syndromes) and the mTOR pathway, which is involved in cellular proliferation, survival, and metabolism. The combination seeks to improve outcomes in patients who have progressed following standard therapies. Both agents are orally administered small molecule drugs, currently in clinical trials in combination for cancers with HIF-pathway dysregulation[1][2][3][4][5][7][8][9][10].
