diazepam + pethidine is a sedation regimen consisting of a benzodiazepine and an opioid analgesic, frequently employed for moderate sedation during endoscopic procedures such as colonoscopy. In the context of North District Hospital studies, the diazepam component is often administered as Diazemuls, a lipid emulsion formulation designed to reduce injection site pain. Diazepam acts as a positive allosteric modulator of the GABA-A receptor, providing anxiolytic, sedative, and amnestic effects. Pethidine (also known as meperidine) serves as a mu-opioid receptor agonist, providing essential analgesia during the procedure. This combination, often referred to as traditional diazemuls-pethidine sedation (DPS), has been used as a standard-of-care comparator in trials evaluating nurse-administered propofol sedation (NAPS).
