Overview

Diaziquone (AZQ, NSC 182986) is an antineoplastic agent with activity against primary nervous system tumors, including gliomas and medulloblastomas. Procarbazine (PCB, NSC 77213) is an alkylating agent used in combination chemotherapy for certain brain tumors. The combination of diaziquone and procarbazine has been studied preclinically in human glioma xenograft models in athymic mice, where it demonstrated enhanced therapeutic effects compared to either drug alone. Specifically, this combination produced significant increases in median growth delay and the number of tumor regressions compared to single-agent therapy[1][2]. The mechanism of action for diaziquone involves DNA alkylation/crosslinking leading to cell cycle arrest and apoptosis; procarbazine also acts as a DNA alkylator but may additionally inhibit protein, RNA, and DNA synthesis[3][4].
