Overview

Didanosine + lamivudine + efavirenz is a three-drug antiretroviral combination used in the treatment of HIV-1 infection. This regimen combines two nucleoside reverse transcriptase inhibitors (NRTIs)—didanosine and lamivudine—with one non-nucleoside reverse transcriptase inhibitor (NNRTI), efavirenz. Didanosine and lamivudine inhibit HIV reverse transcriptase by acting as chain terminators during viral DNA synthesis, while efavirenz binds directly to reverse transcriptase, causing a conformational change that inhibits its activity. This combination has demonstrated efficacy comparable to other first-line regimens for antiretroviral-naive patients with HIV-1 infection[1][2][9]. It is not available as a fixed-dose coformulation but may be prescribed as individual components.
