Overview

This is a four-drug antiretroviral combination composed of **didanosine**, **stavudine**, **nevirapine**, and **emivirine**. Didanosine and stavudine are nucleoside reverse transcriptase inhibitors (NRTIs), and nevirapine is a non-nucleoside reverse transcriptase inhibitor (NNRTI). Emivirine is also a NNRTI, but it is not approved for clinical use and remains an investigational agent. These agents act synergistically to inhibit HIV-1 reverse transcriptase at different binding sites, aiming to suppress viral replication and reduce the progression of HIV infection. Combination antiretroviral therapy is used primarily for HIV-1 infection, but no fixed-dose combination containing these four agents is marketed, and no evidence supports the use of this specific four-drug regimen in clinical practice or guidelines. Didanosine, stavudine, and nevirapine have all been components of historical HIV therapy, while emivirine has not been approved or widely used.
