A combination hormonal contraceptive for men consisting of the synthetic androgen and progestin dimethandrolone undecanoate (DMAU) and the progestin levonorgestrel (LNG). This oral regimen is being investigated for its ability to suppress the hypothalamic-pituitary-gonadal axis, leading to the inhibition of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion, which in turn suppresses spermatogenesis. In this combination, levonorgestrel provides potent progestogenic activity to enhance sperm suppression, while DMAU provides both progestogenic and androgenic activity; the latter is essential to maintain male sexual function and secondary characteristics in the absence of endogenous testosterone. The development of this combination is supported by the National Institute of Child Health and Human Development (NICHD) and has been evaluated in Phase II clinical trials.
