Overview

dimethyl fumarate + interferon beta is an experimental combination therapy for multiple sclerosis (MS) involving two established disease-modifying agents. Dimethyl fumarate is an oral small molecule with anti-inflammatory and neuroprotective effects, primarily via activation of the Nrf2 antioxidant pathway and inhibition of NF-κB, resulting in enhanced cellular defense against oxidative stress and suppression of pro-inflammatory gene expression. Interferon beta is a recombinant protein with immunomodulatory effects, reducing T cell activation and cytokine production. Preclinical and early clinical studies suggest that this combination may have additive or synergistic effects in ameliorating disease activity and protecting neural tissue compared to monotherapy with either agent. The combination has been explored in both animal models and a small clinical context for relapsing-remitting MS, with no major new safety concerns reported.
