Dinaciclib + epirubicin is a combination therapy that was investigated for the treatment of metastatic triple-negative breast cancer (TNBC). Dinaciclib (SCH 727965) is a CDK1/CDK2 inhibitor, while epirubicin is an anthracycline chemotherapy agent. The combination was studied based on preclinical evidence showing that CDK inhibitors combined with anthracyclines were synergistic in decreasing viability of TNBC cell lines. The rationale was particularly relevant for TNBC cases with low molecular weight cyclin E (LMW-E) isoforms overexpression, which occurs in approximately 70% of TNBC cases.
