**Dinutuximab + irinotecan** is a combination therapy comprised of dinutuximab, a chimeric monoclonal antibody targeting disialoganglioside GD2 found on neuroblastoma cells, and irinotecan, a topoisomerase I inhibitor chemotherapeutic agent. Dinutuximab acts through antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity, while irinotecan inhibits DNA replication in cancer cells by preventing the re-ligation of single-strand breaks induced during DNA replication. This combination is being investigated for its antitumor activity primarily in patients with relapsed or refractory neuroblastoma, a pediatric cancer. The regimen is under clinical investigation as an immunochemotherapeutic approach, with the intention that combining targeted immunotherapy with conventional chemotherapy may improve outcomes in neuroblastoma patients[1][4][6].
