This is a combination regimen of three agents used in oncology, particularly for the treatment of GD2-positive cancers such as neuroblastoma and leiomyosarcoma. Dinutuximab beta is a chimeric monoclonal IgG1 antibody that targets the disialoganglioside GD2, which is overexpressed on certain tumor cells including neuroblastoma and some soft tissue sarcomas. By binding to GD2, it induces antibody-dependent cellular cytotoxicity and complement-dependent cytotoxicity, leading to tumor cell apoptosis[1][4][6]. Zoledronic acid is a bisphosphonate that can expand γδ T cells in vivo and has direct anti-tumor effects by inhibiting bone resorption and modulating immune responses[2]. Interleukin-2 (IL-2) is a cytokine that stimulates proliferation and activation of lymphocytes, enhancing immune-mediated anti-tumor activity[3]. The rationale for combining these agents lies in their complementary mechanisms: dinutuximab beta labels tumor cells for immune attack; zoledronic acid expands effector T cell populations; IL-2 further boosts immune response. This combination has been investigated in clinical trials for high-risk neuroblastoma as well as other solid tumors expressing GD2[1][4][6].
