Overview

Diphenoxylate hydrochloride + atropine sulfate is a fixed-dose oral combination medication used to treat diarrhea. Diphenoxylate is an opioid derivative that acts as an antidiarrheal by slowing intestinal motility through agonism of the mu opiate receptor, thereby reducing bowel contractions and prolonging transit time. Atropine sulfate, an anticholinergic agent acting as a muscarinic acetylcholine receptor antagonist, is included in subtherapeutic amounts to discourage abuse by causing unpleasant side effects at higher doses. The combination was approved for medical use in the United States in 1960 and remains widely prescribed for severe diarrhea[1][5][8].
