Overview
Dipyrone (also known as metamizole) combined with codeine is a fixed-dose combination analgesic developed for the treatment of moderate to severe pain, particularly postoperative pain. Dipyrone is a non-opioid analgesic and antipyretic with additional spasmolytic properties. Its mechanism of action involves inhibition of cyclooxygenase enzymes COX-1 and COX-2, leading to reduced prostaglandin synthesis; it may also activate the opioidergic, cannabinoid, and endovanilloid systems. Codeine is an opioid analgesic that acts primarily as a mu-opioid receptor agonist to provide pain relief. The combination aims to enhance analgesia through complementary mechanisms while potentially reducing opioid requirements. Dipyrone/codeine combinations are marketed in some countries but not available in others due to safety concerns regarding dipyrone (notably agranulocytosis). Eurofarma has been identified as a developer for this fixed-dose combination[3][5][1].
