RC48 + PD-1 + platinum-based is an investigational combination regimen consisting of disitamab vedotin (RC48), camrelizumab (a PD-1 inhibitor), and platinum-based chemotherapy. It is being evaluated as a first-line treatment for locally advanced or metastatic HER2-overexpressing (IHC2+/3+) esophageal squamous cell carcinoma (ESCC). Disitamab vedotin is a HER2-directed antibody-drug conjugate (ADC) that delivers the cytotoxic payload monomethyl auristatin E (MMAE) to HER2-expressing tumor cells. Camrelizumab is a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, designed to restore T-cell antitumor activity. Platinum-based chemotherapy agents (such as cisplatin or carboplatin) act as DNA-damaging agents to inhibit tumor cell replication. This triplet regimen aims to leverage the synergistic effects of targeted cytotoxicity, immune checkpoint inhibition, and traditional chemotherapy.
