Overview

Disitamab vedotin + sintilimab is a combination therapy consisting of a HER2-targeted antibody-drug conjugate (disitamab vedotin) and a PD-1 inhibitor (sintilimab). Disitamab vedotin comprises a novel HER2-directed monoclonal antibody (hertuzumab) coupled with monomethyl auristatin E (MMAE) via a cleavable valine-citrulline linker. MMAE is an antimitotic agent that blocks tubulin polymerization, resulting in cell cycle arrest and apoptosis. Sintilimab is a PD-1 inhibitor that enhances T-cell immune responses against cancer cells. This combination is being investigated for HER2-expressing unresectable gastric cancer and gastroesophageal junction adenocarcinoma.
