Overview
Disulfiram + lorazepam is an investigational combination therapy studied by the University of New Mexico for the treatment of alcohol dependence in patients with co-occurring anxiety disorders. Disulfiram, an aldehyde dehydrogenase (ALDH) inhibitor, is used to discourage alcohol consumption by causing an accumulation of acetaldehyde, leading to unpleasant physical reactions. Lorazepam, a benzodiazepine, acts as a positive allosteric modulator of GABA-A receptors to alleviate anxiety symptoms. In this specific clinical approach, lorazepam administration was made contingent on adherence to disulfiram treatment, aiming to improve retention and safety in patients with dual diagnoses. A Phase 4 open-label pilot study (NCT00721526) evaluated the safety and efficacy of this combined regimen.
