divarasib + bevacizumab + FOLFOX is a combination regimen under investigation for the treatment of colorectal cancer and potentially other solid tumors. The regimen combines divarasib, a small molecule inhibitor targeting KRAS G12C, with bevacizumab, a monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), and FOLFOX, a standard chemotherapy regimen consisting of folinic acid (leucovorin), fluorouracil (5-FU), and oxaliplatin. - divarasib (GDC-6036) is designed to selectively inhibit the oncogenic KRAS G12C mutant protein, disrupting downstream signaling that drives tumor growth. - bevacizumab targets and binds VEGF-A, inhibiting angiogenesis required for tumor development and maintenance. - FOLFOX is a cytotoxic chemotherapy regimen that damages DNA (via oxaliplatin) and interferes with DNA/RNA synthesis (via 5-FU and its potentiation by folinic acid)[1][2][3]. This combination is being studied in patients with KRAS G12C-mutated advanced or metastatic colorectal cancer and possibly other KRAS G12C-mutant solid tumors.
