A combination therapy consisting of DKN-01 (sirexatamab), lenalidomide, and dexamethasone, investigated primarily for treatment of relapsed or refractory multiple myeloma. DKN-01 is a humanized IgG4 monoclonal antibody that binds and neutralizes Dickkopf-1 (DKK1), a secreted inhibitor of Wnt signaling associated with tumor growth, immune suppression, and poor prognosis. Lenalidomide is an immunomodulatory small molecule with antineoplastic, antiangiogenic, and pro-immune effects, while dexamethasone is a synthetic corticosteroid with anti-inflammatory and immunosuppressive properties. The combination is designed to suppress tumor proliferation, stimulate immune response, and inhibit tumor-induced immune evasion and bone disease in multiple myeloma.
