This is a combination HIV-1 vaccine regimen composed of three main components: - **SAAVI DNA-C2** (a plasmid DNA vaccine encoding HIV-1 subtype C antigens), - **SAAVI MVA-C** (a recombinant modified vaccinia Ankara vector expressing HIV-1 subtype C antigens), - **gp140 protein** (a V2-deleted clade C envelope glycoprotein from Novartis), adjuvanted with **MF59** (an oil-in-water emulsion adjuvant). The regimen is designed as a prime/boost strategy to induce both cellular and humoral immune responses against HIV. The approach uses sequential or concurrent administration of these components to maximize immunogenicity. Clinical studies in South Africa have shown that this combination is safe and induces neutralizing and binding antibodies as well as CD4+ T-cell responses to the HIV envelope protein. The strongest immune responses were observed when the regimen included an initial prime with either MVA or DNA followed by sequential boosting with gp140/MF59[1][2][3].
