Overview

This is a multi-component experimental HIV vaccine regimen designed for prophylactic use against HIV infection. The combination includes: - **DNA-HIV-PT123**: A DNA vaccine encoding HIV antigens (clade C ZM96 Gag, clade C ZM96 Env, and CN54 Pol-Nef), intended to prime the immune system by inducing cellular and humoral responses. - **CN54gp140**: A recombinant trimeric envelope protein derived from the clade C 97/CN/54 isolate of HIV, used as a protein boost to enhance antibody responses. - **MPLA-L**: Monophosphoryl lipid A in liposomal formulation, an adjuvant that stimulates innate immunity with reduced toxicity compared to native LPS. - **MVA (Modified Vaccinia Ankara-CMDR)**: A non-replicating viral vector engineered to express additional HIV genes (env gp160 CM235 Subtype E and gag/pol CM240 Subtype A), serving as a further boost for both cellular and humoral immunity. The regimen is administered in multiple stages over several months. Its mechanism of action involves priming and boosting both arms of the adaptive immune system—eliciting broadly neutralizing antibodies against diverse HIV strains as well as robust T-cell responses. This approach aims to provide effective prophylaxis against acquisition of HIV[1][3][5][7].
