DNX-2401 + interferon-gamma is an experimental combination therapy for recurrent glioblastoma and gliosarcoma. It consists of DNX-2401, a conditionally replicative, oncolytic adenovirus engineered from human adenovirus serotype 5, which preferentially infects and lyses tumor cells, and interferon-gamma (IFN-γ), a recombinant cytokine that promotes a T-helper 1 (Th1)-skewed immune response. DNX-2401 is administered as a single intra-tumoral injection, followed by subcutaneous IFN-γ initiated two weeks later. This combination is designed to stimulate anti-tumor immunity by direct oncolysis and immune system activation. Clinical trials have shown that while the approach can induce anti-tumor immune responses, tolerability of the IFN-γ component is limited, and no additional benefit from IFN-γ (over DNX-2401 alone) has been definitively established. Key development was driven by academic oncology groups at major cancer centers[1][4][7].
