Docetaxel + bavituximab is a combination therapy investigated primarily for the treatment of advanced non-small cell lung cancer (NSCLC). Docetaxel is a small molecule chemotherapeutic agent in the taxane class that inhibits microtubule depolymerization, thereby disrupting mitosis and leading to apoptosis of rapidly dividing cancer cells[3][5][8]. Bavituximab is a chimeric monoclonal antibody that targets phosphatidylserine, an anionic phospholipid externalized on tumor blood vessels and apoptotic cells in the presence of β2 glycoprotein 1 (β2GP1), exerting antitumor immune effects by promoting immune-mediated destruction of tumor vasculature[1][2][6]. The combination was evaluated in phase III clinical trials for previously treated advanced NSCLC but did not demonstrate superiority over docetaxel alone in terms of overall survival or progression-free survival. Subset analyses suggested possible benefit in patients with high baseline β2GP1 levels or those subsequently receiving immune checkpoint inhibitors[1][2].
