A combination chemotherapy regimen consisting of docetaxel, capecitabine, and cisplatin. Docetaxel is a taxane that promotes the assembly of tubulin into stable microtubules and inhibits their disassembly, leading to a decrease in free tubulin and the inhibition of mitosis. Capecitabine is an orally administered systemic prodrug of 5'-deoxy-5-fluorouridine (5'-DFUR) which is converted to 5-fluorouracil; it inhibits DNA synthesis by blocking the methylation of deoxyuridylic acid to thymidylic acid via thymidylate synthase. Cisplatin is a platinum-based cytotoxic agent that forms DNA adducts, primarily intra-strand cross-links, which inhibit DNA replication and transcription, ultimately leading to programmed cell death. This specific combination was studied by the Roswell Park Cancer Institute in a Phase I trial for patients with advanced or metastatic solid tumors.
