Overview

The combination of docetaxel, cisplatin, and capecitabine—often referred to as the DCX regimen—is a chemotherapy protocol used primarily in the treatment of advanced or resectable gastric cancer and other solid tumors. Docetaxel is a taxane that stabilizes microtubules, preventing their depolymerization and thereby inhibiting mitosis, which leads to apoptosis in cancer cells[6]. Cisplatin is a platinum-based compound that forms DNA crosslinks, interfering with DNA replication and transcription. Capecitabine is an oral prodrug converted to 5-fluorouracil (5-FU) in vivo; it inhibits thymidylate synthase, disrupting DNA synthesis. The triplet regimen has demonstrated significant anti-tumor activity with manageable toxicity profiles and can be used as induction or neoadjuvant therapy for HER2-negative gastric cancer[1][2][4][8]. It may also be considered for other cancers where these agents are individually indicated.
