This is a four-drug combination regimen consisting of docetaxel, cisplatin, cetuximab, and bevacizumab. Docetaxel is a taxane-class chemotherapeutic agent that inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis. Cisplatin is a platinum-based chemotherapy that forms DNA crosslinks and induces apoptosis in rapidly dividing cells. Cetuximab is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling involved in tumor proliferation. Bevacizumab is a humanized monoclonal antibody against vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth. This combination has been investigated primarily as induction therapy for locally advanced or recurrent/metastatic squamous cell carcinoma of the head and neck (HNSCC/SCCHN), aiming to achieve dual inhibition of EGFR and VEGF pathways alongside cytotoxic chemotherapy[1][2]. The regimen has shown feasibility but limited data on efficacy due to small patient numbers in trials[1].
