Overview
Docetaxel + cisplatin + fluorouracil is a chemotherapy combination regimen, commonly referred to as TPF or DCF, used primarily for the treatment of advanced gastric (stomach) cancer and locally advanced squamous cell carcinoma of the head and neck. Each component has a distinct mechanism of action: - Docetaxel (Taxotere) is a taxane that inhibits microtubule depolymerization, disrupting mitosis and leading to cell death. - Cisplatin (Platinol) is a platinum-based compound that forms DNA crosslinks, inhibiting DNA synthesis and function. - Fluorouracil (5-FU) is an antimetabolite that interferes with DNA synthesis by inhibiting thymidylate synthase. The combination exploits different mechanisms to maximize cytotoxicity against cancer cells. This regimen has demonstrated improved response rates in both neoadjuvant and recurrent settings for head and neck cancers compared to doublet regimens[1][2][3][5][6]. All three drugs are FDA-approved for use in these indications[8].
