Overview

This is a multi-agent chemotherapy regimen combining five drugs with distinct mechanisms of action, used primarily in the treatment of advanced or metastatic cancers such as gastroesophageal adenocarcinoma and squamous cell carcinoma of the head and neck. **Docetaxel** is a taxane that inhibits microtubule depolymerization, disrupting mitosis. **Cisplatin** is a platinum-based agent that forms DNA crosslinks, leading to apoptosis. **Fluorouracil** is an antimetabolite that inhibits thymidylate synthase, impairing DNA synthesis. **Leucovorin** enhances the binding of fluorouracil to its target enzyme, increasing efficacy. **Bevacizumab** is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth[1][2]. This combination has been studied in phase II trials for metastatic gastroesophageal cancer and head and neck cancer to improve progression-free survival and response rates over standard regimens.
