The combination of docetaxel, cisplatin, and gemcitabine (often referred to as the GCD regimen) is a multi-agent chemotherapy protocol used primarily for the treatment of advanced or metastatic cancers, including urothelial carcinoma and head and neck cancers. Docetaxel is a taxane that stabilizes microtubules by binding to β-tubulin, thereby inhibiting cell division. Cisplatin is a platinum-based compound that forms DNA crosslinks leading to apoptosis. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporation into DNA and inhibition of ribonucleotide reductase. Together, these agents act synergistically to disrupt cancer cell proliferation through multiple mechanisms targeting both DNA replication and mitosis[1][3][5][6].
