Docetaxel + cisplatin + radiotherapy is a combination regimen used primarily in the treatment of locally advanced non-small-cell lung cancer (LA-NSCLC) and has also been studied in recurrent or incurable squamous cell carcinoma of the head and neck (SCCHN). Docetaxel is a taxane-class chemotherapeutic agent that promotes microtubule assembly and inhibits their disassembly, leading to cell cycle arrest and apoptosis. Cisplatin is a platinum-based chemotherapeutic that forms DNA crosslinks, inhibiting DNA synthesis and function, resulting in apoptosis. Radiotherapy uses ionizing radiation to induce DNA damage directly within tumor cells. The combination aims to maximize cytotoxic effects on cancer cells through complementary mechanisms—chemotherapeutics sensitize tumor cells to radiation-induced damage while targeting different aspects of cellular replication[1][2][3]. This regimen has demonstrated efficacy as an alternative to other platinum-based combinations for LA-NSCLC when given concurrently with thoracic radiotherapy[1][3], offering favorable response rates, survival outcomes, and manageable toxicity profiles.
