The regimen docetaxel + CEF-University of Turku-breast cancer refers to a sequential chemotherapy protocol investigated by the University of Turku in a pharmacogenomics clinical trial (NCT01110291). This treatment consists of three cycles of docetaxel (a taxane) followed by three cycles of the CEF combination, which includes cyclophosphamide (an alkylating agent), epirubicin (an anthracycline), and fluorouracil (an antimetabolite). The study was designed to determine if CYP3A activity and genetic variations in CYP3A5 and MDR1 (ABCB1) could predict the clearance and toxicity of docetaxel in patients with primary high-risk breast cancer. Docetaxel works by stabilizing microtubules and preventing their disassembly, thereby inhibiting mitosis. The CEF components provide complementary mechanisms, including DNA cross-linking, topoisomerase II inhibition, and inhibition of thymidylate synthase, to maximize antineoplastic activity in the adjuvant setting.
