The combination of docetaxel, estramustine, and hydrocortisone is a multi-agent chemotherapy regimen primarily investigated for the treatment of hormone-refractory (androgen-independent) prostate cancer. Docetaxel is a taxane antineoplastic agent that stabilizes microtubules by binding to the β-subunit of tubulin, thereby inhibiting cell division and inducing apoptosis in rapidly dividing cells. Estramustine is an antineoplastic agent combining estradiol with a nitrogen mustard moiety; it acts as both an estrogen receptor agonist and an alkylating agent, causing DNA damage in cancer cells. Hydrocortisone is a corticosteroid used at low doses in this regimen to manage side effects and possibly enhance anti-tumor activity. Clinical studies have shown that this combination produces significant anti-cancer activity with manageable toxicity profiles in men with advanced prostate cancer who have progressed after initial hormone therapy[1][2][3][6][8].
