docetaxel + everolimus

Docetaxel + everolimus is a combination therapy that pairs docetaxel, a taxane chemotherapeutic agent, with everolimus, a mammalian target of rapamycin (mTOR) inhibitor. The combination was investigated for potential synergistic effects in cancer treatment, particularly in non-small cell lung cancer (NSCLC) and breast cancer. Everolimus inhibits the mTOR pathway, which is aberrantly activated in various cancers and known to contribute to resistance associated with taxane therapy. Docetaxel works as an antineoplastic agent. However, clinical trials have shown that this combination is associated with significant toxicities, particularly neutropenia, and variable drug clearance making the combination therapy unpredictable even at low doses.
