The combination of docetaxel, gemcitabine, and imatinib is a triplet regimen that has been studied in phase I clinical trials for the treatment of refractory solid tumors. This combination targets multiple pathways of tumor progression by combining two cytotoxic agents (docetaxel and gemcitabine) with a tyrosine kinase inhibitor (imatinib). Docetaxel: A cytotoxic agent that binds to microtubules, preventing their disassembly and thereby inhibiting cell division. Gemcitabine: A nucleoside analog that inhibits DNA synthesis and induces apoptosis in cancer cells. Imatinib: A tyrosine kinase inhibitor that targets BCR-ABL tyrosine kinase and other kinases, particularly those responsible for uncontrolled cell growth. It works by preventing these enzymes from functioning properly, which slows cancer growth and spread. The combination of these three agents was designed to target multiple pathways simultaneously, potentially enhancing anti-tumor activity.
