Docetaxel + lapatinib is a combination of two antineoplastic agents used primarily in the treatment of HER2-positive metastatic breast cancer. Docetaxel is a taxoid antineoplastic agent that promotes microtubule assembly and stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis in cancer cells. Lapatinib is a small molecule tyrosine kinase inhibitor that targets both epidermal growth factor receptor (EGFR/HER1/ERBB1) and human epidermal growth factor receptor type 2 (HER2/ERBB2), blocking their intracellular phosphorylation domains to prevent downstream signaling involved in tumor cell proliferation and survival. The combination has been studied for its efficacy and tolerability, particularly in patients with advanced or metastatic HER2-positive breast cancer[1][4][6].
