Docetaxel + Low-Dose Abdominal Radiotherapy is a combination therapeutic regimen developed by the Gynecologic Oncology Group (GOG) for the treatment of recurrent, persistent, or advanced ovarian, peritoneal, and fallopian tube cancers. In this approach, low-dose abdominal radiotherapy (LDART) is utilized as a chemosensitizer to potentiate the antineoplastic activity of docetaxel. Docetaxel, a taxane-based chemotherapy agent, works by promoting the assembly of tubulin into stable microtubules and inhibiting their disassembly, which leads to cell cycle arrest in the G2/M phase—a phase where cells are particularly sensitive to ionizing radiation. The concurrent administration of radiation induces DNA damage, which, when combined with the microtubule-stabilizing effects of docetaxel, is intended to overcome resistance and improve clinical outcomes in gynecologic malignancies. This regimen was investigated in a Phase I dose-escalation study to determine the maximum tolerated dose and safety profile.
