Docetaxel + pegylated liposomal doxorubicin is a combination chemotherapy regimen used primarily in the treatment of breast cancer, including as neoadjuvant therapy for patients with axillary lymph node metastasis and metastatic breast cancer. Docetaxel is a taxane that promotes microtubule assembly and inhibits their disassembly, leading to cell cycle arrest and apoptosis. Pegylated liposomal doxorubicin (PLD) is an anthracycline encapsulated in a polyethylene glycol-coated liposome, which prolongs circulation time and reduces cardiotoxicity compared to conventional doxorubicin. The combination leverages the cytotoxic effects of both agents while aiming to minimize overlapping toxicities—particularly cardiac toxicity—by using the pegylated formulation of doxorubicin[1][2][4].
