A combination chemotherapy regimen consisting of docetaxel, a taxoid antineoplastic agent, and S-1, an oral fluoropyrimidine. Docetaxel works by binding to microtubules, stabilizing them and preventing depolymerization, which disrupts cell division and leads to cancer cell death. It also induces apoptosis by binding to the Bcl-2 protein. S-1 consists of tegafur (a prodrug of 5-fluorouracil), a dihydropyrimidine dehydrogenase inhibitor, and potassium oxonate (an orotate phosphoribosyl transferase inhibitor). This combination has shown significant efficacy in treating gastric cancer, with superior 5-year overall survival rates compared to S-1 monotherapy.
