Overview

Docetaxel + sepantronium bromide is a combination therapy consisting of docetaxel, a taxane-based chemotherapeutic agent, and sepantronium bromide (YM155), a small molecule inhibitor of survivin. Docetaxel works by disrupting microtubule function, which is essential for cell division, while sepantronium bromide suppresses survivin expression, a protein that inhibits apoptosis and is overexpressed in many cancer types. The combination has demonstrated synergistic antitumor effects in various cancer models, including ovarian cancer, triple-negative breast cancer, and melanoma. YM155 enhances docetaxel efficacy by downregulating survivin expression, particularly at the G2/M phase of the cell cycle, leading to increased apoptosis compared to either agent alone. Clinical trials have investigated this combination for treating stage III (unresectable) or stage IV melanoma.
