Docetaxel + talabostat is a combination therapy investigated primarily for advanced non-small cell lung cancer (NSCLC) in patients who have failed prior platinum-based chemotherapy. Docetaxel is a taxane-class chemotherapeutic agent that stabilizes microtubules, thereby inhibiting cell division and promoting apoptosis. Talabostat (also known as PT-100) is an orally available amino boronic dipeptide that inhibits dipeptidyl peptidases, including fibroblast activation protein, and enhances immune responses by stimulating cytokine and chemokine production. The rationale for the combination was to leverage both direct cytotoxic effects of docetaxel and the immunomodulatory properties of talabostat to improve antitumor activity. Clinical trials showed that while the regimen was generally well tolerated, there was no significant evidence that adding talabostat enhanced the clinical efficacy of docetaxel in NSCLC[1][2][3].
