This combination therapy consists of docetaxel, a semi-synthetic taxane derivative, and temsirolimus, an mTOR inhibitor. Docetaxel works by binding to and stabilizing microtubules, thereby inhibiting cell division and inducing apoptosis. Temsirolimus is a soluble ester analog of rapamycin that selectively inhibits the Mammalian Target of Rapamycin (mTOR) kinase, a key regulator of cell growth, proliferation, and survival. The combination was evaluated by the Washington University School of Medicine in a Phase I dose-escalation study for patients with resistant solid malignancies, including pancreatic, colorectal, and lung cancers. However, the trial was terminated due to severe myelosuppressive toxicity, and the regimen was not recommended for further clinical development at the tested dose levels and schedules.
