Docetaxel + zanidatamab is a combination therapy under investigation for the treatment of advanced HER2-positive breast cancer. Docetaxel is a well-established taxane-class chemotherapeutic agent that inhibits cell division by stabilizing microtubules, leading to apoptosis in rapidly dividing cells. Zanidatamab (ZW25) is a novel bispecific monoclonal antibody that targets two distinct extracellular domains of the human epidermal growth factor receptor 2 (HER2), resulting in enhanced receptor clustering and internalization, as well as increased immune-mediated cytotoxicity compared to monospecific HER2 antibodies. The combination has demonstrated promising antitumor activity and a manageable safety profile as first-line therapy for patients with advanced HER2-positive breast cancer in phase 1b/2 clinical trials[1][2][3][4][5][6][7].
