NeoCART is a Phase 2, multicenter, randomized, open-label clinical trial (NCT03154749) conducted by Guangdong Provincial People's Hospital to compare two neoadjuvant chemotherapy regimens for early-stage triple-negative breast cancer (TNBC). The experimental regimen consists of six cycles of docetaxel and carboplatin (DCb), while the standard control regimen consists of four cycles of epirubicin and cyclophosphamide (EC) followed by four cycles of docetaxel (D). Docetaxel is a taxane that stabilizes microtubules, carboplatin is a platinum agent that induces DNA cross-linking, epirubicin is an anthracycline that intercalates DNA and inhibits topoisomerase II, and cyclophosphamide is an alkylating agent. The trial demonstrated that the carboplatin-based regimen (DCb) achieved a significantly higher pathologic complete response (pCR) rate of 61.4% compared to 38.6% for the anthracycline-based regimen (EC-D) in patients with stage II or III TNBC.
