Dolutegravir + RTI refers to the use of dolutegravir in combination with a reverse transcriptase inhibitor (RTI), which is standard in antiretroviral therapy for HIV-1 infection. Dolutegravir is an HIV-1 integrase strand transfer inhibitor (INSTI) that blocks the integration of viral DNA into the host genome by inhibiting the strand transfer step mediated by HIV integrase. This action prevents viral replication within human cells. Dolutegravir is known for its high genetic barrier to resistance and favorable pharmacokinetic profile, allowing once-daily dosing without food restrictions or pharmacokinetic boosting[3][5]. It is used in both treatment-naïve and treatment-experienced patients and can be co-formulated with other antiretrovirals such as lamivudine or abacavir[5]. The addition of an RTI (such as lamivudine or abacavir) provides a complementary mechanism—blocking reverse transcription of viral RNA into DNA—resulting in potent suppression of HIV replication[1][5].
