Overview

Domperidone + rivastigmine is a combination of two pharmaceutical agents used together, primarily in the management of Alzheimer's disease or Parkinson's disease dementia. Rivastigmine is a cholinesterase inhibitor that works by inhibiting both acetylcholinesterase and butyrylcholinesterase, thereby increasing acetylcholine levels in the brain to improve cognitive function. Domperidone is a peripheral dopamine D2 receptor antagonist used as an antiemetic; it does not cross the blood-brain barrier significantly and helps prevent gastrointestinal side effects such as nausea and vomiting commonly associated with cholinesterase inhibitors like rivastigmine. This combination allows for higher dosing or improved tolerability of rivastigmine by mitigating its dose-limiting gastrointestinal adverse effects[2][1][4].
